Darkfield Microscopes
Used in dark field microscopy, the appropriately-named darkfield microscope illuminates its specimens in such a way as to reduce or almost completely reduce the amount of superfluous light to exclude all but the most important parts of the observable image. The result is an image with a high level of contrast between the sample and the rest of the image, typically represented as the field surrounding the sample being dark with the sample itself appearing bright.
Dark field microscopy has a number of advantages. Constructing a darkfield microscope is often simple, as they begin life as simple compound microscopes and are then fitted with darkfield condensers and specialized shields in front of the light source to focus the light exclusively on the sample. Applications for dark field microscopy include observing live biological samples safely without having to resort to staining them and killing them. Darkfield microscopes are also used in metallurgy and gemology to look for imperfections and otherwise hard-to-spot elements that would be otherwise impossible to spot with more traditional bright field microscopes.
